This presentation describes a preliminary study on equivalency of PLS/PCA NIR models built using SIMCA® compared to current validated models developed using different software. Test models were built in SIMCA® using same calibration spectra as current models, with similar spectra pre-treatment. Equivalence was assessed by primary outputs (scores/loadings), specific model predictions and outlier diagnostics. This effort is expected to allow manufacturing sites flexibility to use different multivariate software for operability/business reasons.

Traditional process monitoring relies on univariate plots, making it labour-intensive and often leading to post-process analysis. In contrast, multivariate data analysis condenses multiple plots into key visualizations, enabling Operators to quickly identify trends and anomalies. This approach serves as an early warning system, enhancing real-time monitoring, improving productivity, and reducing the risk of costly deviations.

Continuous cell culture offers high productivity for biologics like monoclonal antibodies but presents challenges in sustaining cell density and productivity over time. In this study, PAT tools integrated with the Ambr 250 HT system enabled automated monitoring and control of critical culture parameters. Results showed that low Biomass Specific Perfusion Rates (BSPRs) boosted cell productivity, demonstrating the power of PAT-driven, adaptive control for enhancing bioprocess outcomes.
